Animal models are invaluable for modelling genetic disorders and to find treatments for diseases. The mouse is the foremost vertebrate experimental model because its genome has been sequenced completely and can be precisely and variously engineered.

Recombineering (Red/ET recombination) has become the method of choice to engineer large replicons, such as targeting constructs (large high-copy plasmids, low-copy plasmids) or bacterial artificial chromosomes (BACs). Such large constructs are essential to subsequently prepare models with sufficient significance. Since recombineering can be performed at any position in the DNA the technology has opened up an unlimited number of possibilities for even complex genetic modifications.

Gene Bridges has over 10 years experience and know-how from over 500 DNA engineering projects. Due to the advantages of recombineering for the generation of targeting constructs, which bypasses multiple laborious and mutagenic EtBr and UV preparative steps, our targeting constructs recombine in ES cells with remarkable efficiencies, saving time and money at this expensive step!

Costumers only name the target gene and describe the desired modification while our service team takes care of the rest!

  • decent timeline and regular progress updates

  • final construct confirmed by restriction analysis and sequencing

  • purified and linearized DNA (on request)

  • graphical Southern screening strategy (targeting constructs only)

We can offer you the preparation of:

  • conditional knock-out targeting constructs

  • knock-in targeting constructs (e.g. point mutations, humanization)

  • conditional knock-in targeting constructs

  • reporter targeting constructs (or BAC transgenes)

  • FLP/Cre/CreERT2/Dre expressing targeting constructs (or BAC transgenes)

  • removal of “passenger genes” from BAC clone (“BAC shaving”)
